All: Oh!
Phoebe: Please!
Monica: No! No unscheduled stops. You can go when we stop for gas.
Chandler: Oh, come on, there's a rest stop right up there! Come on, I really have to goooooooooo.
"I have to go," is a very childish way to express that you need to use the bathroom. So it's cute. Chandler is saying it sarcastically 4. He knows how childish he is being.
Joey: Oh, now I have to go!
Since Joey is the most immature 5 and childish out of the whole group, it makes sense for him to say it seriously.
